Neoplasms in Purebred Boxer Dogs following Long-Term Administration of A/-Methyl-A/-nitrosourea1

چکیده
The carcinogenicity of N-methyl-N-nitrosourea (MNU) has been demonstrated in several species for a variety of tissues, particularly the nervous system. Boxer dogs, a breed with a high incidence of spontaneous tumors (in cluding brain tumors), were exposed to MNU to determine their susceptibility to MNU and to compare the induced tumors with the types of tumors reported in the literature. Twenty purebred Boxer dogs were given weekly i.v. injections of MNU, 5 mg/kg, for 36 weeks and were observed for approximately 3 years for the development of neoplasia. Nineteen of 20 dogs developed neoplasms 19 to 36.5 months after the first injection. The mean latent period was 30 ±5 (S.D.) months. All tumor-bearing dogs had 2 or more neoplasms, including métastases, and 14 dogs had tumors of more than 1 tumor type. Malignant neurinomas of the small intestine were present in 11 dogs, and poorly differentiated sarcomas of the small intestine were found in 6 dogs. Metastasis occurred to liver, spleen, lymph nodes, lung, and adrenal. Two cases of malignant neurinoma developed in the heart, and 1 each developed in the stomach and colon. Two welldifferentiated neurinomas occurred in the small intestine, and 1 each occurred in a spinal root, the stomach, and the colon. The high incidence rate, young age of development, and unique tumor types clearly distinguished the tumors from spontaneous tumors of Boxers. Although the lack of brain involvement was unexplained, MNU had a neurooncogenic effect on the peripheral nerves of the gastrointes tinal tract.

متن کاملPrevalence of Dog Erythrocyte Antigen 1 in 7,414 Dogs in Italy
The study aim was to establish the prevalence of DEA 1, the most immunogenic and clinically important blood group in canine blood transfusion, in 7,414 dogs from Italy. The potential sensitization risk following a first transfusion and the acute reaction risk following a second transfusion given without a cross-matching and blood typing test were also calculated. Dogs tested were purebred (4,79...
متن کاملEstimating canine cancer incidence: findings from a population-based tumour registry in northwestern Italy
BACKGROUND Canine cancer registry data can be put to good use in epidemiological studies. Quantitative comparison of tumour types may reveal unusual cancer frequencies, providing directions for research and generation of hypotheses of cancer causation in a specific area, and suggest leads for identifying risk factors. Here we report canine cancer incidence rates calculated from a population-bas...
متن کاملQuinapril Attenuates the Effect of Long-Term L-NAME Administration on the Vascular Reactivity of Diabetic Rats
Angiotensin-converting enzyme (ACE) inhibitors including quinapril could exert a protective effect on cardiovascular system through endothelial system in normoglycemic and diabetic rats. The present experimental work was designed to study the vascular reactivity of aortic ring segments isolated from streptozotocin (STZ)-diabetic rats treated for 4 weeks with nitro-L-arginine-methyl ester (L-NAM...
متن کاملThe Study of Histomorphometry and Histochemistry of Liver in Adult Male and Female Rats Following Short-term and Long-term Administration of Methylphenidate
Background & Objective: Methylphenidate is one of the most common medications used for treatment of attention-deficit/hyperactivity disorder (ADHD). The administration of methylphenidate, through blocking of dopamine receptors and increment of extracellular dopamine levels, leads to escalation in central nervous system activity. Numerous studies investigated the effect of methylphenidate on bod...
